Influence of the streptozotocin on the corticotrope axis of the Wistar rat (Rattus norvegicus)

Résumé

Ce travail présente les altérations de paramètres biologiques et histologiques de l'axe cortico-surrénalien pendant 1, 2 et 3 semaines chez le rat Wistar rendu diabétique insulinodépendant suite à l'administration intrapéritonéale de streptozotocine (STZ) à raison de 45mg/kg de poids corporel. Le travail a été axé sur l’étude histophysiologique de la glande surrénale, appuyé par un dosage de certains paramètres plasmatiques (glycémie, triglycéridémie, cholestérolémie et ACTHémie plasmatique) chez les rats traités par rapport aux rats témoins. Au vu des résultats obtenus, il ressort que la streptozotocine entraîne l’installation du syndrome diabétique qui se caractérise par une polyphagie, une polyurie, une polydipsie et une glycosurie. Ce syndrome diabétique se traduit aussi par une chute du poids corporel couplée à un désordre métabolique affectant la glycémie, la triglycéridémie ainsi que la cholestérolémie. L'évolution des différents paramètres biologiques est transitoire car les variations observées retrouvent le niveau basal observé avant l'administration de STZ. Une nette augmentation du taux plasmatique d’ACTH s’opère à la deuxième semaine suivie d’une baisse progressive pour rejoindre le taux basal en fin d’expérimentation. En outre, l’étude histologique de la glande surrénale révèle l’existence de corrélations entre l’état physiologique de l’animal et les modifications structurales et fonctionnelles de cette glande après traitement par la STZ. On assiste également à une migration de la zone médullaire vers la glomérulée et à l’installation d’une fibrose d’intensité progressive signalant ainsi l’état pathologique de l’animal. Le rétablissement de l’état normal en fin d’expérimentation chez les animaux sous STZ nous laisse supposer que la dose de STZ administrée à raison de 45 mg/kg de poids corporel semble être insuffisante pour maintenir l’état diabétique.

Abstract

This work presents the alterations of biological and histological parameters of cortico- adrenal axis during 1, 2 and 3 weeks in Wistar rats made insulin-dependent diabetic following the intraperitoneal administration of streptozotocin (STZ) at a rate of 45mg/kg of body weight. Work was centered on the histophysiologic study of the adrenal gland, supported by a proportioning of certain plasmatic parameters (glycemia, triglyceridemy, cholesterolemy and plasmatic ACTHemy) in the rats treated compared to the control rats. Considering the results obtained, it appears that the streptozotocine involves the installation of the diabetic syndrome which is characterized by a polyphagy, a polyury, a polydipsy and a glycosury. This diabetic syndrome also results in a fall of the body weight coupled to a metabolic disorder affecting the glycaemia, the triglyceridemy as well as the cholesterolemy. The evolution of biological parameters is transient because the variations observed find the basal level prior to administration of STZ. A clear increase in the plasmatic rate of ACTH takes place at the second week of experimentation followed by a progressive fall to join the basal rate at the end of the experimentation. Moreover, the histological study of the adrenal gland reveals the existence of correlations between the physiological state of the animal and the structural and functional modifications of this gland after treatment by the STZ. We also attend with a proliferation of the medullar zone in glomerulosa and the installation of a fibrosis of progressive intensity, thus announcing the pathological state of the animal. The re-establishment of the normal state at the end of the experimentation in the animals under STZ lets us suppose that the amount of STZ managed at a rate of 45 mg/kg of body weight seems to be insufficient to maintain the diabetic state.
